Patient‑Specific Profiling of TCR‑T Cell Efficacy Enabled by a Rapid and Standardized Reporter System

  • TCR-T cell therapy has demonstrated significant clinical potential, yet its efficacy is often compromised by tumor-mediated suppression of antigen presentation. Current screening methods, primarily based on genomic and transcriptomic sequencing, fail to fully capture physiological pHLA abundance or direct T-cell responsiveness. Here, we developed a standardized and robust reporter system that recapitulates TCR-T cytotoxic activity both in vitro and in vivo. This platform enables rapid, patient-specific assessment of therapeutic potential using ex vivo tissues within 12 hours. By providing a direct functional readout, this system offers substantial translational utility for early patient stratification and the evaluation of TCR-T cell therapies.
